Evaluation of HLA-G and IL10 expression levels in HCV infected patients receiving combination therapy

Abstract :

Hepatitis C disease is virus mediated infection causing major health problem worldwide. Conversions of immune surveillance play important roles in viral clearance. Immune modulating molecules such as HLA-G and IL-10 that shift immune response to T helper 2 (Th2) may play a role to inhibit response to combination therapy with IFN-α and Ribavirin. The aim of this study was to investigate the expression of HLA-G and IL-10 in HCV infected patients. In this study, HCV viral load and genotype were assessed using an in-house real-time RT-PCR assay. The expression levels of HLA-G and IL-10 were also evaluated by a developed real-time PCR method. Moreover, soluble HLA-G in both groups of patients (48 responder and non-responder) and 50 healthy individuals were determined by enzyme-linked immunosorbent assay (ELISA). According to obtained data, HCV genotype 1a was predominant in both responder and non-responder patients. Expression levels of HLA-G and IL-10 in non-responder group was significantly more that responder and control groups (P <0.001). Also, expression levels of HLA-G and IL-10 in non-responder at the beginning of treatment was remarkably higher in comparison with healthy individuals. After treatment, soluble HLA-G level was significantly increased in non-responders when compared to responders (P <0.001). These findings suggest that elevated expression level of HLA-G and IL-10 in HCV infected patients play an important role in response to combination therapy.

She is scientific member of Lorestan University and research fellow in Digestive Disease research Institute (DDRI). She is also the director of reference lab in Health deputy of Lorestan University of Medical Science and the manager of talented organization in Lorestan Province.
