Expression of NF-κB pathway in peripheral blood monocytes of incipient psoriasis patients

Abstract :

Nuclear Factor Kappa B (NF-κB) is a nuclear transcription factor with regulation of cytokines, adhesion molecules and chemokines gene expression in skin tissue, even involved in skin inflammatory disease process. The study found that NF-κB was highly expressed in the skin lesion of psoriasis patients, suggesting that the occurrence of psoriasis was related to NF-κB pathway. But nobody knows whether it is also expressed in the blood especially mononuclear cell. We enrolled 20 healthy persons and 30 patients with incipient psoriasis patients, extracted their peripheral blood and collected their Peripheral Blood Mononuclear Cells (PMBC) by density gradient centrifugation. Western blot was used to detect the changes of NF-κB related protein (p50, p65, IKKβ, IκBα, p-IKBα and p-p65) in PMBC. The results showed that the expression levels of p50, p65, IKKβ, IκBα, p-IKBα and p-p65 on peripheral blood mononuclear cells of incipient psoriasis patients were significantly increased in psoriasis group compared with control group. It is concluded that the NF-κB pathway plays an important role in the pathogenesis of psoriasis.
