HCV is a leading health issue of Pakistan, number of HCV genotypes and subtypes circulating across Pakistan. The key question of this study was to figure out the frequency of s HCV genotypes and subtype among the Pakistani HCV isolate. A total of 6464 HCV positive serum samples collected the across the country were tested by genotyping assay. Out of 6464 HCV RNA positive patients, 3704 were from Punjab, 1036 Khyber Pakhtunkhwa, 767 Sindh, 90 Balochistan, 596 Islamabad, 258 Kashmir, 9 FATA and 4 Gilgit-Baltistan. A total 6464 tested serum samples by type-specific PCR fragments, sample distribution of genotypes of was as follows: 230 genotype 1; 2 genotype 2; 5755 genotype 3, mixed genotype; 53 genotype (3a+1a); 235 genotype (3a+1b); 4 genotype (3a+2a);3 genotype (3a+2b); 182 genotype (3a+3b). The finding of subtype was as 158 1a, 72 1b, 1 2b, 5389 3a, mixed genotype 1(2a+3a), 53 (3a+1a), 235 (3a+1b), 4 (3a+2a), 3 (3a+2b), 181 (3a+2 3a), 364 3b, 1(3a+1b) and 2 3c. The prevalent genotype in Pakistan is 3a (89.03%), with the highest number in the age groups 21-30 (1268), 31-40 (1797) and 41-50 (1249).
