Abstract :

Lab-scale production of exoglucanase by Escherichia coli containing the respective gene from Thermotoga petrophila was studied using 3-liter fermenter. Different fermentation media were used to analyze the expression of exoglucanase out of which terrific broth comprising 2.4% yeast extract, 1.2% tryptone, 72 mM K2HPO4, 17 mM KH2PO4 and 0.4% glycerol was found to be the most suitable for the production of exoglucanase, giving an activity of 8.10 U/mg after 24 hours of incubation. Optimum incubation temperature, medium pH, concentration of IPTG and inoculum size for maximum activity of exoglucanase were found as 37Ë?C (9.32 U/mg), 6.0 (11.08 U/mg), 0.75 mM (12.79 U/mg) and 1% (12.83 U/mg), respectively. In order to obtain maximal production of exoglucanase, effect of different carbon sources, organic and inorganic nitrogen sources was also studied. 1.0% of lactose (12.99 U/ mg) as a carbon source, 1.0% of meat extract (13.19 U/mg) as an organic nitrogen source and 1.0% of ammonium chloride (13.34 U/ mg) as an inorganic nitrogen source were evaluated to be the most suitable. Fermentation kinetics was also applied for the effective validation of the results.
